My company wants me to relocate, to another country.  Because of my family, I can't.

I started looking for a new job, and haven't said no to my boss yet. 

How and when do I tell him?  There will be no job for me here if I don't relocate.

How well does your boss know you? Should he not already be aware of the fact that you can not take the offer to relocate? If you have a good contact, he already knows, and I think you can only win his help by talking to him about your situation.

My boss is overseas.  He is a good boss, but he sticks to business.  So he doesn't know my situation. 
So I don't know what to say to him.

I'm not seeing an upside to telling him.  If you had a healthy manager/direct relationship you would want to nurture that as primary value, but since it sounds like that doesn't exist, I'm not seeing an immediate benefit to informing him.  Follow the process outlined in the How To Resign podcast to ensure you depart professionally.
If, however, you're considering another position with your current employer, you'll do best to tell him now.
